PLASMA MEMBRANE DOMAINS IN INTERNODAL CELLS OF THE CHARACEAN GREEN ALGAE: PHOTOSYNTHESIS, ION CURRENTS, MEMBRANE POTENTIAL AND PROTEOMICS

    The pH along the surface of internodal cells and photosynthesis will be
    measured with micro pHelectrodes and pulse-amplitude modulated
    microfluorometry; confocal scanning laser microscopy will be used to
    visualize charasomes together with fluorescent Reporters for H+, Ca+,
    Cl-; extracellular ion fluxes will be measured using vibrating electrodes;
    electrophysiological methods will be used to detect lateral differences
    in plasma membrane potential; mass spectrometry methods should
    reveal differences in protein composition between charasome-rich and
    charasome-poor areas

    The overall goal of this research project is (1) to correlate ion fluxes,
    photosynthetic activity and, eventually, lateral differences in plasma
    membrane potential and intracellular ion concentration with the
    distribution of convoluted plasma membrane domains of characean
    green algae in vivo by live cell imaging and (2) to identify charasome
    specific proteins by proteomics approaches. According to the current
    hypotheses, hitherto based on indirect evidence, the charasomes are
    responsible for proton extrusion (pH-banding), required for effective
    carbon acquisition and photosynthesis, and play an important role in
    Cl- uptake and Ca2+ transport. So far, however, nothing is known
    about the protein composition of charasomes apart from an
    uncharacterized proteoglucan which may be responsible for
    membrane curvature. We will therefore apply mass spectrometry
    analysis in order to identify proteins specifically enriched in or absent
    from charasomes in comparison with smooth plasma membrane areas.
