Description
The aim of this project is to quantitatively investigate the inhibition of RNA-protein binding by small molecules identified by Angara’s Chemoproteomics platform using Fluorescence Correlation spectroscopy (2D-FIDA). This will include the establishment of a 2D-FIDA binding assay for interaction of the RNA binding protein SRSF1 with its canonical binding motifs. This assay will then be used to quantitatively investigate the inhibition of SRSF1-RNA binding by up to 24 small molecules provided by Angara to derive IC50 values. In addition, this will allow to evaluate the application potential of Fluorescence Correlation Spectroscopy (ie 2D-FIDA) as a miniaturised platform technology for stratification of RNA binding protein inhibitors.
