Disclosing the Biodiversity and Evolutionary Relationships of Planktonic Ciliates (Alveolata, Ciliophora, Oligotrichea) Based on Big Data from the Northwest Pacific Biodiversity Hotspot and the North Atlantic

    Project Details

Plankton samples will be taken from coastal sites and the open sea of the northern China Seas to the South China Sea, and cultures shall be established to provide enough material for species descriptions, investigations of the cell divisions, and ultrastructural studies. Water samples for high-throughput sequencing shall be collected from both Chinese coastal sites and the open sea and European coastal waters at different seasons. Species descriptions and redescriptions will be based on barcoding combined with live observations and studies of protargol-impregnated material. The ultrastructure of the species is investigated by means of scanning and transmission electron microscopy. The genetic and morphologic data are analysed to infer the phylogenetic relationships among the Oligotrichea. For biogeographical studies, the V4 region of the small subunit ribosomal RNA is analysed in the plankton material by high-throughput sequencing, and sequence similarity networks are employed.

The main proposal objectives are (i) descriptions of Oligotrichea from the China Seas at highest scientific level including barcoding and cell division, (ii) ultrastructural studies in the northwest Pacific endemites and taxa relevant for the proof of evolutionary hypotheses, (iii) performing cladistic and phylogenetic analyses, and (iv) comparing the biodiversity of Oligotrichea in different oceans and testing the hypothesis of dispersal limitation in planktonic ciliates from the China Seas, using high-throughput sequencing and sequence similarity networks.
